病理病態学生体反応病理学(病理学第一)

概要

酸化ストレスの生物学的意義を細胞からヒト個体レベルまで広くかつ深く追究しています。病理学固有の形態学を重視しながら、遺伝学的・細胞工学的・遺伝子工学的な手法を駆使して以下のテーマに取り組んでいます。出身分野にかかわらず、酸化ストレスに深い興味を持つ学生が研究室に参加することを望みます。当講座では、遺伝子改変動物の解析に必須の形態学的な知識を広く身につけることが出来ます。MDのひとは、希望により病理医としての基礎的な研修を併行して行うことができます(名古屋大学医学部附属病院としての病理専門医研修プラグラムに組み込まれています)。関連病院での非常勤勤務も可能です。教室を主宰する豊國伸哉は日本酸化ストレス学会ならびに日本鉄バイオサイエンス学会の理事長であり、国際的にはSociety for Free Radical Research InternationalのPresidentです。

研究プロジェクト

1.活性酸素による発がんメカニズムの解析

25年に渡り当研究室で解析されてきた鉄ニトリロ三酢酸による腎発がんモデルの解析もいよいよ佳境に入ってきています。2002年2月にはAm J Pathol誌に、主な標的遺伝子のひとつであるp16のallelic lossが投与開始後数週のたいへん早い時期に起こっていることをCommentary付きで発表しました。いよいよヒト発がんへの応用、全ゲノム的な解析を開始しています。また、最近ではオキシゲノミクス(Oxygenomics)というコンセプトを提唱し、ゲノム内で酸化的傷害の起こりやすい部位を網羅的に解析することに取り組んでいます。オキシゲノミクスを総説で解説しています(Toyokuni S and Akatsuka S, J Clin Biochem Nutr 39: 3-10, 2006)。2006年秋にはオキシゲノミクスの方法論に関する論文を発表しました。このモデルのCGH解析により、酸化ストレスによりwild-type動物に発生したがんではそのゲノムの変化がヒトのがんに極めて類似することが判明しました(Akatsuka et al. PLoS ONE 2012)。これは、ヒトのがんの根本原因も酸化ストレスにあることを示唆しています。がんからゲノムの酸化的な傷害へ仕事は大きく進展しています。また、このモデルを使用したがん予防法の検討にも取り組んでいます。

2.繊維状無機物質の毒性・発がん性の研究

project2.jpg平成18年度よりラボをあげて、アスベスト病態の解明に取り組んでいます。鉄含有率の高いアスベスト繊維ほど、発がん性の高いことが疫学的に明らかにされていますが、わたしたちの研究により鉄を含んでいない白石綿(クリソタイル)も体内で鉄を集めることにより、極めて発がん性が高いことが明らかとなっています(Jiang et al. J Pathol 2012)。鉄発がんの研究からこれまでに蓄積してきたノウハウを結集して、サイエンスから社会への還元を目指して取り組んでおり、除鉄に予防効果のあることがわかりつつあり(Nagai et al. Cancer Prev Res 2013)、新規治療薬の開発にも取り組んでいます。また、最近では、新規素材として期待されているカーボンナノチューブの毒性・発がん性評価にも社会からの要請から取り組んでいます。その結果、50 nm程度の太さの多層カーボンナノチューブにはアスベストと同様の中皮腫発がん性があることが判明し、この種の素材は注意して扱うべき事を警告しました(Nagai et al. Proc Natl Acad Sci USA 2011)。この結果はIARCにより50 nmカーボンナノチューブのGroup 2Bの根拠にも使用されています。現在のところ、固形物内で使用されており、アスベストと違い、一般消費者に害が及ぶ可能性は少ないと考えています。現在、この発がんメカニズムについても詳細に検討中です。

3.酸化ストレスマーカーの開発、酸化的組織細胞傷害の可視化に関する研究

project3.jpg上記1および2より得られた成果をもとに、新たな酸化ストレスマーカーの開発を行っています。酸化ストレスは,発がんのみならず動脈硬化症・糖尿病などのさまざまな生活習慣病の病態に関わっていることがわかってきており、そのモニタリングは今後ますます重要になると考えられます。最近では、フェントン反応の基点となる触媒性二価鉄を可視化する方法を共同研究として開発しています(Mukaide et al. Free Radic Res 2014)。

4.低温プラズマの医療応用に関する研究

project4.jpg平成24年より医工連携としてこのプロジェクトに取り組んでいます。プラズマとは固体・液体・気体に次ぐ第4の物理的状態で、気体・電子・ラジカル・イオン・紫外線などを混じた状態です。太陽や雷が自然界に存在するプラズマですがいずれも高温です。ところが、90年代から体温に近い低温プラズマの作成が可能となりました。この医療応用をめざしています。これまでの研究により、低温プラズマにより種々の程度の酸化ストレスを意図的に与えることができることが明らかになってきており(Okazaki et al. J Clin Biochem Nutr 2014)、今後、殺菌・止血やがん治療など幅広い応用が可能であると考えられます。
