OUTLINE
神経細胞に特有のカルシウムシグナリング(細胞内のカルシウム濃度変化とそれに伴い活性化される一連の分子経路)は大変多彩で、記憶や情動変化など健康な 脳機能発揮において大変重要な役割を果たすことが分かっています。その裏腹に、カルシウムシグナリングの異常が精神疾患の発症に寄与することが示唆されて いますが、その病態はほとんど分かっていません。
当研究室では、神経細胞に特徴的なカルシウムシグナリングという分子経路を手掛かりに、脳機能 の成立と破綻のしくみを明らかとすることを目指しております。具体的には、どのように、①脳が作られるか、②変化するか、③病気になるか、3つの異なる生 理、病理現象について、分子細胞基盤を明らかとする研究を推進します。上記に対し、分子細胞生物学、遺伝学、組織学、行動学、回路操作、in vivoを含むカルシウムイメージングなど、新旧様々な技術を駆使し、主にマウスをモデル動物として用いた基礎研究を行います。
神経科学の研究分野では、近年目覚ましい技術革新があり、これまで知り得なかったことに取組むことができるようになってきました。機能不明のカルシウム依存性蛋白質リン酸化酵素 (CaMKIγ/CL3)の全長を発見したことをきっかけに、2000年に開始した私たちの研究も、当初は主に培養神経細胞の研究を行っておりましたが、今日では脳内の神経回路を研究の対象とすることが多くなりました。
また、一方で、ゲノム解読技術の急速な発展に伴い、ヒトを含む多くの生物における全ゲノム配列が明らかにされ、精神神経疾患の病態への寄与が示唆されるゲ ノム変異知見の蓄積が一層進んでおります。これに伴い、臨床研究によって見出されたゲノム変異知見を、基礎研究により意義付ける研究が益々重要になると考 えます。このような、臨床知見を起点として、基礎研究による分子病態理解を推進し、医学の発展へと貢献する研究にも、注力していきたいと考えます。
新しい技術が次々と開発され、今後益々の展開が期待される神経科学はとても面白く、難しくもやりがいがあります。当研究室では、上記の研究を推進することにより、最終的には、生命現象における真理の追究と、医学・医療への貢献を目指します。
